Summary
Curi Bio is refining its established tissue casting process by introducing the Mantarray Mini Plates, designed to enhance cost efficiency by reducing tissue size while maintaining robust data. This miniaturization not only lowers cell consumption and increases cell density, but also enables new applications while preserving the high functionality and >95% casting success rate Mantarray users are familiar with. We highlight the applicability of this advancement through the development of a new Cardiac Media formulation, specifically optimized for human 3D engineered heart tissue experiments. This innovative formula is designed to more accurately model in vivo cardiac development and maturation, through inclusion of numerous metabolites found in human plasma that are essential for cardiac function at physiological concentrations.
In this webinar, Curi Bio will present an overview of the Mantarray 3D contractility instrument and detailed overview of the mini casting plates and their benefits. Functional assessment of mini 3D engineered heart tissues (EHTs) will compare and contrast contractility and drug responses in conventional RPMI-based media and physiologically relevant Curi Bio media designed to support oxidative metabolism and improved maturation of muscle constructs. Cardiac organoids cultured in Curi Bio medium contract with greater force, faster kinetics, and remain functionally active for extended culture times, allowing for long term disease modeling, therapeutic studies, and toxicity screens. Notably, drug responses are more accurate and reliable when EHTs are cultured in Curi Bio medium suggesting improved translatability of preclinical safety and efficacy screening. Available in comprehensive growth and maintenance kits, this media supports every stage of in vitro cardiac muscle tissue development in 3D platforms.

Dr. Luttrell is Product Manager for Cell and Tissue Models at Curi Bio. She is leveraging her stem cell and bioengineering expertise to lead the development and commercialization of cell and media consumables. She plays a pivotal role on the Product Management team, supporting and building customer relationships, while integrating research and product development to increase the pre-clinical utility of in vitro cell-based assays.
